Reiterated transgene is detectable by DNA in situ hybridisation. Congenic 129/terSv-TgN(Hbb-b1)83Clo,Pax6<Sey/+>/Ws mice carry the TgN(Hbb-b1)83Clo transgene on a 129/terSvHsd genetic background and are segregating for Pax6<Sey/+> and Pax6<+/+>. The transgene is a useful lineage marker for chimera studies. ES cells are most easily derived from the 129 strain. The 129/terSv-TgN(Hbb-b1)83Clo,Pax6<Sey/+>/Ws congenic strain should be useful for producing Pax6<Sey/+> and Pax6<+/+> ES cells with the TgN(Hbb-b1)83Clo lineage marker. These ES cells will be useful in production and analysis of Pax6<Sey/+> Pax6<+/+>, Pax6<Sey/Sey> Pax6<+/+> and other chimeras.
Females homozygous for Tg(hbb-b1)83Clo and heterozygous for Pax6 were mated to 129/terSvHsd males and then backcrossed to 129/terSvHsd until N10. N10 offspring were intercrossed to generate mice homozygous for the transgene and segregating for Pax6 and Pax6<+/+>. The homozygous stock are maintained by intercrossing. For embryo freezing, 129/terSvHsd females are crossed to 129/terSv-Tg(Hbb1-b1)83Clo, Pax6/Ws. Therefore the embryos should be hemizygous for the transgene and 50% should be Pax6.
